I'd start with Perdido Street Station [Amazon Link].

If from there you decide to read the rest of his books that take place in the same universe (No, its not really a trilagy. Characters in subsequent books know about things that happened in previous books, but each book is a discrete story.) I'd read the Scar next (good) then I'd pick up a copy of his short stories -- Looking for Jack -- because there is a cool story in there about Jack Half-a-Prayer who makes a cameo in Perdido Street Station and is important as history to Iron Council which is one of the best books I've ever read.

There are other good stories in Looking for Jake. I really liked the Tain.

King Rat is an earlier Mielville effort that I liked and is a bit different from his more recent novels. More of a contemporary London fairey-tale gone dark and horror movie.
